Circular Dichroism of Light-harvesting Complexes from Purple Photosynthetic Bacteria*

چکیده
The CD spectra of a range of antenna complexes from several different species of purple photosynthetic bacteria were recorded in the wavelength range of 190 to 930 nm. Analysis of the far UV CD (190 to 250 nm) showed that in each case except for the B800-850 from Chr. vinosum the secondary structure of the light-harvesting complexes contains a large amount of a-helix ( 2 5 0 % ) and very little p-pleated sheet. This confirms the predictions of the group of Zuber of a high a-helical content based upon consideration of the primary structures of several antenna apoproteins. The CD spectra from the carotenoids and the bacteriochlorophylls show considerable variations depending upon the type of antenna complex. The different amplitude ratios in the CD spectrum for the bacteriochlorophyll Qy, Qx and Soret bands indicate not only different degrees of exciton coupling, but also a strong and variable hyperchromism (Scherz and Parson, 1984a, b).

The peripheral light-harvesting complexes from purple sulfur bacteria have different 'ring' sizes.
The integral membrane light-harvesting (LH) proteins from purple photosynthetic bacteria form circular oligomers of an elementary unit that is composed of two very hydrophobic polypeptides, termed alpha and beta. These apoprotein dimers are known to associate into closed circular arrays of 8, 9 and 16 alpha/beta-mers. We report the existence of peripheral LH proteins purified from Allochromatiu...

Spectroscopy of individual light-harvesting 2 complexes from purple photosynthetic bacteria revealed a deformation of the circular complex into C(2) symmetry. The present work relates the geometry of the deformed aggregate to its spectroscopic properties. Different models of elliptical deformation are discussed and compared with the experimental findings. It is shown that the model with smaller...
متن کاملThe minimal model of light harvesting complex with dipole-quadrupole interaction derived from ab initio calculation
A minimal, one-parameter model of the excitonic Hamiltonian for the light harvesting complex of purple bacteria (LH2) based on ab initio calculation of the excitonic energies is proposed. The set of input parameters contains positions of atoms in 27 bacteriochlorophyll pigments only. The excitation energies, transition dipole and quadrupole moments of the bacteriochlorophyll units were calculat...

Absorption and circular dichroism (CD) spectra of light-harvesting (LH)1 complexes from the purple bacteria Rhodobacter (Rba.) sphaeroides and Rhodospirillum (Rsp.) rubrum are presented. The complexes exhibit very low intensity, highly nonconservative, near-infrared (NIR) CD spectra. Absorption and CD spectra from several mutant and reconstituted LH1 complexes, with the carotenoid neurosporene ...
متن کاملN-terminal methylation of the core light-harvesting complex in purple photosynthetic bacteria.
Several core light-harvesting complexes from both sulfur and non-sulfur purple photosynthetic bacteria have been identified to be methylated at the N-terminal alpha-amino group of beta-polypeptides by using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and nuclear magnetic resonance. Monomethylation has been confirmed for the N-terminal alanine residues of the bet...
